It would be very difficult to take this statement at face value, since (as explained on page 31) תכלת was available throughout the times of the אמוראים and was still in use by the רבנן סבוראי and perhaps the גאונים . This means that for 500-800 years after the
Churban Bayis, תכלת was still being used.
One possible explanation of the Arizal’s statement may be that the earliest decrees against תכלת began around the time of the Churban (see page 31), foreshadowing the impending disappearance of this מצוה . It is not saying that precisely on the day of the Churban it disappeared. If this is correct, then it is certainly possible that תכלת may very well return in the centuries preceding the end of the Galus. In our time, we are only slightly more than 200 years away from the end of the .אלף השישי
In any case, one must always be wary of quoting anything from the
Arizal out of context, since these statements require much
background information to be understood properly [158]. One thing is
certain; the Arizal surely did not intend to uproot a מצוה from the
Torah, at a time that it is possible to fulfill.[159]

Item
Ari’s writings reflect spiritual decline, not a prohibition. Tekheiles use continued centuries after Churban Bayis Sheni. There’s no halachic basis to reject a mitzvah when Hashem restores the means.
